Известно, что рецепторы горького вкуса (TAS2R) экспрессированы на многих клетках респираторной системы, а их активация сопровождается широким спектром эффектов, потенциально полезных для терапии бронхиальной астмы (БА).</p></sec><sec><title>Цель</title><p>Цель. Оценить влияние полиморфизмов генов TAS2R3, TAS2R4 и TAS2R5 на формирование БА, показатели вентиляционной функции легких и контроль заболевания.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В исследование было включено 240 больных БА различной тяжести (возраст 43,9±1,03 года, 44,5% мужчин) и 90 здоровых добровольцев (контрольная группа, возраст 38,0±1,09 лет, 50% мужчин). Функцию внешнего дыхания оценивали методом стандартной спирометрии, контроль заболевания определяли с помощью вопросника ACT. В результате предварительного отбора по частоте минорного аллеля, прогнозирования функциональной значимости и оценки неравновесия по сцеплению, полиморфизм TAS2R4 rs33920115 был определен в качестве репрезентативного варианта для группы полиморфизмов TAS2R3, TAS2R4 и TAS2R5. Генотипирование выполняли методом ПЦР с анализом плавления ампликонов в высоком разрешении.</p></sec><sec><title>Результаты</title><p>Результаты. Полиморфизм rs33920115 был значимо ассоциирован с БА в кодоминантной (p=0,01), доминантной (p=0,006), рецессивной (p=0,03), лог-аддитивной (p=0,003) и мультипликативной (p=0,003) моделях наследования. Носительство генотипа AA чаще отмечалось среди больных БА (29,2% против 17,8% в контрольной группе), тогда как гомозиготы GG были чаще представлены в группе контроля (33,3% против 19,2%). Эффект оставался значимым после коррекции на пол и возраст (ОШ 1,8; 95%ДИ (1,26-2,61), p=0,001 для лог-аддитивной модели). Мы не обнаружили влияния rs33920115 на вентиляционную функцию легких и контроль БА.</p></sec><sec><title>Заключение</title><p>Заключение. Полиморфизм TAS2R4 rs33920115 и связанные с ним вариации генов TAS2R3 и TAS2R5 могут влиять на предрасположенность к развитию БА, вероятно, за счет изменения экспрессии соответствующих рецепторов.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. It is known that bitter taste receptors (TAS2R) are expressed on many cells of the respiratory system and their activation is accompanied by a wide range of potentially beneficial effects for the treatment of asthma.</p></sec><sec><title>Aim</title><p>Aim. To evaluate the influence of TAS2R3, TAS2R4 and TAS2R5 polymorphisms on the development of asthma, parameters of lung function and control of the disease.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The study enrolled 240 patients with asthma of varying severity (mean age 43.9±1.03 years, 44.5% males) and 90 healthy volunteers (control group, mean age 38.0±1.09 years, 50% males). Lung function was assessed using standard spirometry, the disease control was determined by ACT questionnaire. Preliminary screening by minor allele frequency, prediction of functional significance and assessment of linkage disequilibrium allowed us to select TAS2R4 rs33920115 as a representative variant for the group of TAS2R3, TAS2R4 and TAS2R5 polymorphisms. Genotyping was performed by PCR with high-resolution melting analysis.</p></sec><sec><title>Results</title><p>Results. rs33920115 polymorphism was significantly associated with asthma in codominant (p=0.01), dominant (p=0.006), recessive (p=0.03), log-additive (p=0.003) and multiplicative (p=0.003) genetic models. Carriage of the AA genotype was more often observed among patients with asthma (29.2% vs. 17.8%) while the GG homozygotes were prevalent in the control group (33.3% vs. 19.2%). The effect remained significant after adjustment for sex and age (OR 1.8; 95%CI (1.26-2.61), p=0.001 for log-additive model). We found no effect of rs33920115 on lung function and asthma control.</p></sec><sec><title>Conclusion</title><p>Conclusion.
